serum. The maximum cell concentration was a function of the frequency of medium renewal, the time of incubation, and the amount of final cellular material present. Maximum cell density could be increased approximately twofold by the addition of various peptones to the complete medium, although these materials did not affect the minimum generation time. Fortification of the medium with glucose, glutamine, or vitamin mixture did not prolong the growth phase or affect the generation time. When complete medium was incubated for 8 days at 37° C., it was found to be fully

An improved tissue culture assay. III. Alternate methods for measuring cell growth.
Simplified methods were described for the measurement of growth of human car cinoma cells in tissue culture: (a) determination of color intensity in lysates of cells stained with crystal violet (CV method), (6) determination of the reduction of 2,6dichlorophenol indophenol, and (c) determination of total organic solids (dichromate method). The three methods gave comparable measurements of relat...
متن کاملAn Improved Tissue Culture Assay I. Methodology and Cytotoxicity of Anti-Tumor Agents*
A technic for the determination of cytotoxieity of chemical compounds for mammalian cells in vitro has been described by Eagle and Foley (~). Their assay requires 7 days for completion, with renewal of medium every other day. An application of the Eagle-Foley technic, in which the total assay time is 5 days with medium changes at 1 and 3 days, has been worked out with fermentation products (1~)...
متن کاملGrowth suppression effect of human mesenchymal stem cells from bone marrow, adipose tissue, and Wharton's jelly of umbilical cord on PBMCs
Objective(s):Immunosuppressive property of mesenchymal stem cells (MSCs) has great attraction in regenerative medicine especially when dealing with tissue damage involving immune reactions. The most attractive tissue sources of human MSCs used in clinical applications are bone marrow (BM), adipose tissue (AT), and Wharton's jelly (WJ) of human umbilical cord. The current study has compared immu...
متن کاملAn improved tissue culture assay. II. Cytotoxicity studies with antibiotics, chemicals, and solvents.
In the preceding publication of this series (10), a simplified tissue culture assay was described. The cytotoxic activity of various nucleic acid antagonists, antibiotics, amino acid and vitamin antagonists, respiratory poisons, miscellaneous chemicals, solvents, heavy metals, detergents, and fermentation beers were determined with the new into sterile bottles and dissolved in water (sterile), ...
متن کاملI-16: Assessment of The Vitrified Ovarian Tissue in Long Term Culture
In vitro culture of human ovarian tissue the following cryopreservation is proposed for follicular development. There are no techniques that guarantee successful maturation of the follicles within the excised tissue. The viability of cultured human ovarian tissue improved by adding some growth factors to the culture media. The efficiency of vitrification as the cryopreservation method for human...
